Optimization of Growth Conditions to Identify the Superior <i>Bacillus</i> Strain Which Produce High Yield of Thermostable Alpha Amylase

Thermostable α-amylases hold a very important place in commercial industrial applications Sri Lanka. Therefore, the main aim of this study was to identify superior Bacillus strain and optimize growth conditions that could yield high α-amylase production. Three strains, B. amyloliquefaciens ATCC 23350, licheniformis 14580 megaterium 14581 were used for study. Shake flask culture experiments conducted effect various fermentation such as temperature, incubation period, carbon source, nitrogen initial pH concentration on extracellular DNSA assay carried out determine enzyme activity. The highest temperature activity reported by at 85°C, followed 75°C 45°C. Both able give their optimum production 37°C, while 30°C 150 rpm with 7. gave after 48 h 24 incubation. Among sources tested cassava starch For amyloliquefaciens, obtained 2% starch, tryptone source Maximum 1.5% KNO3 6. 1% 7.5 induced According results obtained, is thermostable alpha amylase producer. However, according requirement, can also be an producer due its stability higher temperatures.
